Abstract
This invention relates to isolated nucleic acids encoding plant defensins. The invention also relates to the construction of a chimeric gene encoding all or a portion of the plant defensin, in sense or antisense orientation, wherein expression of the chimeric gene results in production of altered levels of plant defensins in a transformed host cell.
Claims
- THAT WHICH IS CLAIMED:
- An isolated nucleic acid molecule comprising a nucleotide sequence selected from the group consisting of:
- (a) a nucleotide sequence set forth in SEQ ID NO: 29, 31, 34, 46, or 48;
- (b) a nucleotide sequence that encodes a polypeptide comprising the amino acid sequence set forth in SEQ ID NO: 32, 35, 47, or 49;
- (c) a nucleotide sequence characterized by at least 90% sequence identity to the nucleotide sequence set forth in SEQ ID NO: 31, 34, or 48;
- (d) a nucleotide sequence characterized by at least 96% sequence identity to the nucleotide sequence set forth in SEQ ID NO: 46;
- (e) a nucleotide sequence that encodes a polypeptide comprising an amino acid sequence having at least 93% sequence identity to the amino acid sequence set forth in SEQ JD NO: 47;
- (f) a nucleotide sequence that comprises the complement of any one of (a) to (e).
- The nucleic acid molecule according to claim 1, wherein the nucleotide sequence is optimized for expression in a plant.
- A DNA construct comprising a nucleotide sequence of claim 1 , wherein said nucleotide sequence is operably linked to a promoter that drives expression in a host cell.
- An expression cassette comprising the DNA construct of claim 3.
- A host cell having stably incorporated into its genome at least one DNA construct of claim 3.
- The host cell of claim 5, wherein said host cell is a plant cell.
- A plant having stably incorporated into its genome the DNA construct of claim 3.
- The plant according to claim 7, wherein said plant is a monocot.
- The plant according to claim 7, wherein said plant is a dicot.
- Transformed seed of the plant of claim 7.
- An isolated polypeptide selected from the group consisting of:
- (a) a polypeptide comprising the amino acid sequence set forth in SEQ JD NO: 32, 35, 47, or 49;
- (b) a polypeptide comprising an amino acid sequence having at least 90%) sequence identity to the amino acid sequence set forth in SEQ ID NO: 35; and
- (c) a polypeptide comprising an amino acid sequence having at least 93%o sequence identity to the amino acid sequence set forth in SEQ JD NO: 47.
- The polypeptide of claim 11, wherein said polypeptide is characterized by defensin-like activity against at least one plant pathogen.
- The polypeptide of claim 12, wherein said plant pathogen is a fungus.
- The polypeptide of claim 13, wherein said fungus is Sclerotinia sclerotiorum.
- The polypeptide of claim 11 , wherein said polypeptide is characterized by pesticidal actiyity against at least one insect pest.
- The polypeptide of claim 11 , wherein said polypeptide is characterized by pesticidal activity against at least one nematode.
- The polypeptide of claim 16, where said nematode is the Soybean Cyst Nematode.
- A method for impacting a plant pathogen comprising introducing into a plant or cell thereof at least one nucleotide construct comprising a nucleotide sequence operably linked to a promoter that drives expression of a coding sequence in plant cells, wherein said nucleotide sequence is selected from the group consisting of:
- (a) the nucleotide sequence set forth in SEQ ED NO: 29, 31 , 34, 46, or 48;
- (b) a nucleotide sequence that encodes a polypeptide comprising the amino acid sequence set forth in SEQ JD NO: 32, 35, 47, or 49;
- (c) a nucleotide sequence characterized by at least 90% sequence identity to the nucleotide sequence set forth in SEQ ED NO: 31, 34, or 48;
- (d) a nucleotide sequence characterized by at least 96% sequence identity to the nucleotide sequence set forth in SEQ ED NO: 46;
- (e) a nucleotide sequence that encodes a polypeptide comprising an amino acid sequence having at least 93% sequence identity to the amino acid sequence set forth in SEQ JD NO: 47;
- (f) a nucleotide sequence that comprises the complement of any one of (a) to (e).
- The method of claim 18, wherein said plant produces a polypeptide characterized by defensin-like activity against at least one pathogen.
- The method of claim 19, wherein said pathogen is a fungus.

- The method of claim 20, wherein said fungus is Sclerotinia sclerotiorum.
- The method of claim 18, wherein said plant produces a polypeptide characterized by pesticidal activity against at least one insect pest.
- The method of claim 18, wherein said plant produces a polypeptide characterized by pestcidal activity against at least one nematode.
- The method of claim 23, wherein said nematode is a Soybean Cyst Nematode.
- A method for enhancing resistance of a plant to at least one fungal pathogen comprising introducing into a plant or cell thereof at least one nucleotide construct comprising a nucleotide sequence operably linked to a promoter that drives expression of said nucleotide sequence in plant cells, wherein said nucleotide sequence encodes a polypeptide that has defensin-like activity against said at least one fungal pathogen, wherein said at least one fungal pathogen is Sclerotinia sclerotiorum.
- The method of claim 25, wherein said nucleotide sequence comprises a sequence having at least 90%> identity to the coding region set forth in SEQ JD NO: 31, 34, or 48.
- The method of claim 25, wherein said nucleotide sequence comprises a sequence having at least 96%> identity to the coding region set forth in SEQ ED NO: 46.
- The method of claim 25, wherein said nucleotide sequence comprises the coding region set forth in SEQ JD NO: 46.
- A method for enhancing resistance of a plant to at least one plant pathogen comprising introducing into a plant or cell thereof at least one nucleotide construct comprising a nucleotide sequence operably linked to a promoter that drives expression of said nucleotide sequence in plant cells, wherein said nucleotide sequence encodes a polypeptide that has defensin-like activity against said at least one plant pathogen, wherein said at least one plant pathogen is a nematode.
- The method of claim 28, wherein said nematode is the Soybean Cyst Nematode.
- The method of claim 28 wherein said nucleotide sequence comprises a sequence having at least 90% identity to the coding region set forth in SEQ JD NO: 31, 34, or 48.
- The method of claim 28, wherein said nucleotide sequence comprises a sequence having at least 96% identity to the coding region set forth in SEQ JD NO: 46.
- The method of claim 28, wherein said nucleotide sequence comprises the coding region set forth in SEQ JD NO: 46.
