However, they are lack of inhibition activity on PKC and Rho kinase, leading to resistance to pulmonary artery relaxation. KMUP-1 and KMUP-3 (10-8 to 10-4M) reduced U46619-induced pulmonary hypertension and plasma oxygen consumption, comparable to milrilone and zaprinast. They further increased eNOS expression and reduced U46619-induced contraction and Rho kinase expression, similar to Y27632, in isolated rat pulmonary artery preparations. The ability of KMUP-1 and KMUP-3 on U46619-induced pulmonary artery contractions were reduced by pretreatements with PKC activator PMA (1x10-5M), sGC inhibitor ODQ (1.0 μM), endothelium nitric oxdide synthase (eNOS) inhibitor L-NAME (100 μM), and adenylate cyclase inhibitor SQ 22536 (10 μM). KMUP-1 increased more plasma cGMP and KMUP-3 increased more cAMP after their intravenous injection, both of them increased the plasma oxygen concentration. It is suggested that relaxation effects by KMUP-1 and KMUP-3 on pulmonary artery might be mediated by the stimulation of eNOS/cGMP and AC/cAMP, respectively. Both situation were accompanied with inhibition of Rho kinase and protein kinase C (PKC) expression.
